I am studying a Cys2His2 zinc finger protein called NRSF/REST (neuron restrictive silencer factor/RE-1 silencing transcription factor). NRSF/REST is a biologically important transcriptional repressor that uses a cluster of eight zinc fingers to bind its 21bp binding site.
Outside the lab, my husband and I busy ourselves by wrangling our boy/girl twins — every day is an adventure, both in the lab and at home! |

I am working with Morgan to use zinc finger nucleases to modify genes in human cells. I am also interested in using the OPEN protocol to create zinc finger nucleases with novel specificities followed by a quantitative determination of the specificities and affinities of the engineered products as measured in vitro.

My current focus is on engineering zinc finger proteins to bind target DNA sequences in endogenous genes using the OPEN (Oligomerized Pool ENgineering) method recently developed in the lab. I am working on using these proteins as a tool for gene targeting in ES and iPS cells.
When I am not in the lab, I enjoy baking, camping, traveling to new places and reading Harry Potter.

I'm primarily interested in zinc finger nuclease technology as it applies to gene therapy. My projects are targeted at increasing gene targeting efficiency and reducing cytotoxicity of zinc finger nucleases. I am also working to develop methods to examine the genomewide effects of zinc finger nucleases. |
